No. There might be a little grease or transmission oil there when you take it off but it won't drain the transmission.

Yes, if the speedo gear of the vehicle you are asking about is on the ****** it is likely to leak ****** fluid when removed. On this site however we mostly discuss Model A Fords and on those the speedo drive is mounted on the driveshaft torque tube and will not, of course, drain the ******.
